There is water in my quartz sleeve. Will this affect my UV?

Yes, there should be no water in the quartz sleeve.  You will want to make sure that the quartz sleeve has no cracks in it and that the socket is pushed about a 1/4" inside of the quartz sleeve.  Water in the quartz sleeve can cause the lamp and ballast to go out premature.

You recommend water testing the UV before putting it into use on my aquarium-what does that mean?

This means that you will want to connect a pump to the UV to push water through it to check for leaks-most people do this in a sink or with a bucket, but be sure not to submerge the UV under water.
